    A study of ASRS reports involving general aviation and weather encounters

    Consideration is given to the nature and characteristics of problems involving dissemination of weather information, use of this information by pilots, its adequacy for the purpose intended, the ability of the air traffic control system to cope with weather related incidents, and the various aspects of pilot behavior, aircraft equipment, and NAVAIDS affecting flights in which weather figures. It is concluded from the study that skill and training deficiencies of general aviation pilots are not major factors in weather related occurrences, nor is lack of aircraft equipment. Major problem causes are identified with timely and easily interpreted weather information, judgement and attitude factors of pilots, and the functioning of the air traffic control system

    The role of advection, straining, and mixing on the tidal variability of estuarine stratification

These observations result from three additional processes not accounted for in the traditional tidal straining model: 1) along-channel and 2) lateral advection of horizontal gradients in the vertical salinity gradient and 3) tidal asymmetries in the strength of vertical mixing. As a result, cross-sectionally averaged values of the vertical salinity gradient are shown to increase during the flood tide and decrease during the ebb. Only over a limited portion of the cross section does the observed stratification increase during the ebb and decrease during the flood. These observations highlight the three-dimensional nature of estuarine flows and demonstrate that lateral circulation provides an alternate mechanism that allows for the exchange of materials between surface and bottom waters, even when direct turbulent mixing through the pycnocline is prohibited by strong stratification.The funding for this research was obtained from NSF Grant OCE-08-25226.2012-11-0

    Broadband acoustic quantification of stratified turbulence

The definitive version was published in Journal of the Acoustical Society of America 134 (2013): 40-54, doi:10.1121/1.4807780.High-frequency broadband acoustic scattering techniques have enabled the remote, high-resolution imaging and quantification of highly salt-stratified turbulence in an estuary. Turbulent salinity spectra in the stratified shear layer have been measured acoustically and by in situ turbulence sensors. The acoustic frequencies used span 120ā€“600 kHz, which, for the highly stratified and dynamic estuarine environment, correspond to wavenumbers in the viscous-convective subrange (500ā€“2500 māˆ’1). The acoustically measured spectral levels are in close agreement with spectral levels measured with closely co-located micro-conductivity probes. The acoustically measured spectral shapes allow discrimination between scattering dominated by turbulent salinity microstructure and suspended sediments or swim-bladdered fish, the two primary sources of scattering observed in the estuary in addition to turbulent salinity microstructure. The direct comparison of salinity spectra inferred acoustically and by the in situ turbulence sensors provides a test of both the acoustic scattering model and the quantitative skill of acoustical remote sensing of turbulence dissipation in a strongly sheared and salt-stratified estuary.This work was supported by NSF grant OCE- 0824871, ONR grant N00014-0810495, and WHOI internal funds

    The influence of stratification and nonlocal turbulent production on estuarine turbulence : an assessment of turbulence closure with field observations

The definitive version was published in Journal of Physical Oceanography 41 (2011): 166-185, doi:10.1175/2010JPO4470.1.Field observations of turbulent kinetic energy (TKE), dissipation rate Īµ, and turbulent length scale demonstrate the impact of both density stratification and nonlocal turbulent production on turbulent momentum flux. The data were collected in a highly stratified salt wedge estuary using the Mobile Array for Sensing Turbulence (MAST). Estimates of the dominant length scale of turbulent motions obtained from the vertical velocity spectra provide field confirmation of the theoretical limitation imposed by either the distance to the boundary or the Ozmidov scale, whichever is smaller. Under boundary-limited conditions, anisotropy generally increases with increasing shear and decreased distance to the boundary. Under Ozmidov-limited conditions, anisotropy increases rapidly when the gradient Richardson number exceeds 0.25. Both boundary-limited and Ozmidov-limited conditions demonstrate significant deviations from a local productionā€“dissipation balance that are largely consistent with simple scaling relationships for the vertical divergence in TKE flux. Both the impact of stratification and deviation from equilibrium turbulence observed in the data are largely consistent with commonly used turbulence closure models that employ ā€œnonequilibriumā€ stability functions. The data compare most favorably with the nonequilibrium version of the L. H. Kantha and C. A. Clayson stability functions.     Neuroinflammation and J2 Prostaglandins: Linking Impairment of the Ubiquitin-Proteasome Pathway and Mitochondria to Neurodegeneration

    The immune response of the CNS is a defense mechanism activated upon injury to initiate repair mechanisms while chronic over-activation of the CNS immune system (termed neuroinflammation) may exacerbate injury. The latter is implicated in a variety of neurological and neurodegenerative disorders such as Alzheimer and Parkinson diseases, amyotrophic lateral sclerosis, multiple sclerosis, traumatic brain injury, HIV dementia, and prion diseases. Cyclooxygenases (COX-1 and COX-2), which are key enzymes in the conversion of arachidonic acid into bioactive prostanoids, play a central role in the inflammatory cascade. J2 prostaglandins are endogenous toxic products of cyclooxygenases, and because their levels are significantly increased upon brain injury, they are actively involved in neuronal dysfunction induced by pro-inflammatory stimuli. In this review, we highlight the mechanisms by which J2 prostaglandins (1) exert their actions, (2) potentially contribute to the transition from acute to chronic inflammation and to the spreading of neuropathology, (3) disturb the ubiquitin-proteasome pathway and mitochondrial function, and (4) contribute to neurodegenerative disorders such as Alzheimer and Parkinson diseases, and amyotrophic lateral sclerosis, as well as stroke, traumatic brain injury (TBI), and demyelination in Krabbe disease. We conclude by discussing the therapeutic potential of targeting the J2 prostaglandin pathway to prevent/delay neurodegeneration associated with neuroinflammation. In this context, we suggest a shift from the traditional view that cyclooxygenases are the most appropriate targets to treat neuroinflammation, to the notion that J2 prostaglandin pathways and other neurotoxic prostaglandins downstream from cyclooxygenases, would offer significant benefits as more effective therapeutic targets to treat chronic neurodegenerative diseases, while minimizing adverse side effects

    Feminization of Male Brown Treesnake Methyl Ketone Expression via Steroid Hormone Manipulation

    Pheromones are useful tools for the management of invasive invertebrates, but have proven less successful in field applications for invasive vertebrates. The brown treesnake, Boiga irregularis, is an invasive predator that has fundamentally altered the ecology of Guam. The development of control tools to manage Boiga remains ongoing. Skin-based, lipophilic pheromone components facilitate mating in brown treesnakes, with females producing the same long-chain, saturated and monounsaturated (ketomonoene) methyl ketones known to function as pheromones in garter snakes, Thamnophis sirtalis. Boiga also express novel, diunsaturated methyl ketones (ketodienes) with a purported function as a sex pheromone. In our study, we implanted 17 Ī²- estradiol in adult male brown treesnakes in order to manipulate methyl ketone expression as sex attractants, an effect that would mirror findings with garter snakes. Specifically, estrogen promoted production of two ketomonoenes, pentatriaconten-2-one and hexatriaconten-2-one, and suppressed production of one ketodiene, heptatriacontadien-2-one. In bioassays, estrogen-implanted males elicited tongue-flicking and chin rubbing behavior from unmanipulated males, though the responses were weaker than those elicited by females. On Guam, wild males exhibited greatest responses to whole female skin lipid extracts and only weak responses to the methyl ketone fractions from females and implanted males. Our results suggest that sex identity in brown treesnakes may be conferred by the ratio of ketomonoenes (female) to ketodienes (male) from skin lipids and may be augmented by a sex-specific endocrine signal (estradiol). However, a blend of long-chain methyl ketones alone is not sufficient to elicit maximal reproductive behaviors in male Boiga

    The role of knowledge structures in fault diagnosis

    The use of human memory and knowledge structures to direct fault diagnosis performance was investigated. The performances of 20 pilots with instrument flight ratings were studied in a fault diagnosis task. The pilots were read a scenario which described flight conditions under which the symptoms which are indicative of a problem were detected. They were asked to think out loud as they requested and interpreted various pieces of information to diagnose the cause of the problem. Only 11 of the 20 pilots successfully diagnosed the problem. Pilot performance on this fault diagnosis task was modeled in the use of domain specific knowledge organized in a frame system. Eighteen frames, with a common structure, were necessary to account for the data from all twenty subjects

    ChIPing the cistrome of PXR in mouse liver

    The pregnane X receptor (PXR) is a key regulator of xenobiotic metabolism and disposition in liver. However, little is known about the PXR DNA-binding signatures in vivo, or how PXR regulates novel direct targets on a genome-wide scale. Therefore, we generated a roadmap of hepatic PXR bindings in the entire mouse genome [chromatin immunoprecipitation (ChIP)-Seq]. The most frequent PXR DNA-binding motif is the AGTTCA-like direct repeat with a 4bp spacer [direct repeat (DR)-4)]. Surprisingly, there are also high motif occurrences with spacers of a periodicity of 5ā€‰bp, forming a novel DR-(5nā€‰+ā€‰4) pattern for PXR binding. PXR-binding overlaps with the epigenetic mark for gene activation (histone-H3K4-di-methylation), but not with epigenetic marks for gene suppression (DNA methylation or histone-H3K27-tri-methylation) (ChIP-on-chip). After administering a PXR agonist, changes in mRNA of most PXR-direct target genes correlate with increased PXR binding. Specifically, increased PXR binding triggers the trans-activation of critical drug-metabolizing enzymes and transporters. The mRNA induction of these genes is absent in PXR-null mice. The current work provides the first in vivo evidence of PXR DNA-binding signatures in the mouse genome, paving the path for predicting and further understanding the multifaceted roles of PXR in liver

    Addressing secondary traumatic stress, burnout, resilience and turnover in the child welfare workforce: Results from a 6-month, cluster-randomized control trial of Resilience Alliance

    Introduction: US child welfare agencies have historically struggled with workforce retention and turnover. As part of the Quality Improvement Center for Workforce Development in Child Welfare, we tested an adaptation of the Resilience Alliance (RA) model in a large, Midwestern state to address workplace stress, burnout and actual workforce turnover. RA is a 24-week, facilitated program designed to mitigate the impact of secondary traumatic stress among child welfare professionals, and to therefore increase job satisfaction, resilience and optimism and to decrease turnover, stress reactivity and burnout. Methods: Supervisory units of caseworkers and supervisors were randomized to the RA treatment condition (n = 192) or a control condition (no intervention; n = 183). Hypothesis: We hypothesized that participation in the RA adaptation would cause the workforce to experience lower levels of secondary traumatic stress (STS), burnout and intent to search for work or leave their current position. We hypothesized that RA would lead to higher reported levels of resilience and intent to stay. Furthermore, if hypothesized changes were observed due to participation in RA, then such participation would also lead to decreased actual workforce turnover over a 2.5-year period. Results: There were no statistically significant effects of the intervention on changes in STS, burnout or resilience between treatment and control groups over a 6-month period. Participation in RA did cause significant differences in 6-month changes for four turnover intention measures. Finally, RA had no statistically significant effect on turnover.
